High-throughput screening for natural compound-based autophagy modulators reveals novel chemotherapeutic mode of action for arzanol. マウス抗ヒトWIPI2抗体、クローン2A2は、WIPI49様タンパク質2としても知られるWDリピートドメインホスホイノシチド相互作用タンパク質2（WIPI-2）を認識します。WIPI2は、3つのWDリピートを含む454アミノ酸〜54kDaのオートファゴソームマーカーです。 WIPI2は、酵母タンパク質Atg18の哺乳類オルソログであり、同様に初期のオートファゴソーム構造に動員され、成熟したオートファゴソームへの成熟に必要です（Polson et al.2010）。 ヒトWIPI2は、WIPI2A、標準的な454アミノ酸アイソフォームおよびWIPI2Bを含む複数のアイソフォームで存在し、N末端領域とC末端領域の両方に欠失があります。 マウス抗ヒトWIPI2抗体であるクローン2A2は、C末端配列を使用して生成され、ウエスタンブロッティングによってWIPI2AとWIPI2Bの両方を認識します（Pantoom et al.2020）。 マウス抗ヒトWIPI2抗体であるクローン2A2は、ヒト網膜上皮細胞株RPE1におけるWIPI2の免疫蛍光検出に使用されています（MacVicar and Lane2014）。 |
